Bacterial transformation experiment  को frederick griffith ने 1928 ईस्वी में सिद्ध किए थे । अपने experiment के लिए उन्होंने  streptococcus pneumoniae  नामक जीवाणु को चुना और जब उस जीवाणु पर अध्ययन( search) किए तो उन्होंने दो प्रकार का जीवाणु पाया, दोनो में अलग-अलग strain  पाया। उन्होंने एक बैक्टीरिया का पूरा शरीर की कोशिका के भक्ति के ऊपर शैलेशमा युक्त आवरण से घिरा हुआ था जो चिकना था  Griffith ने उसे S- type का बैक्टीरिया नाम दिया। और दूसरे बैक्टीरिया का कोशिका भित्ति किसी शैलेशमा  युक्त आवरण से नहीं घिरा  था अत: वह खुरदरा था जिसे वैज्ञानिक ने R- type का बैक्टीरिया माना। अब Frederick Griffith ने S- type के जीवाणु को चूहे के अंदर inject किया जिससे चूहे को निमोनिया हो गया। क्योंकि यह जीवाणु निमोनिया रोग भी पैदा करते हैं और दूसरे चूहा में R- type के जीवाणु inject किया जिससे चूहे को निमोनिया नहीं हुआ। अत: यह नहीं मारा। जबकि S- type वाले  बैक्टीरिया से चूहा मर गया था। जिससे चूहा मरा उसे virulent तथा जिससे नहीं मरा उसे nanvirulent  जीवाणु कहा गया।

अब S- type के जीवाणु को उच्च ताप देकर मार दिया गया और मृत अवस्था में ही R- type के जीवाणु के साथ मिश्रण  किया गया और उस  मिश्रण को एक तीसरे चूहे में inject किया गया जिससे चूहा मर गया। ऐसा इसलिए हुआ  क्योंकि R- type के जीवाणु में मरे हुए S- type  के जीवाणु का DNA transfer कर गया था। अतः वैज्ञानिक ने इस प्रयोग को bacterial transfer नाम दिया साथ ही साथ इस प्रयोग से यह भी स्पष्ट हो गया कि सचमुच DNA एक अनुवांशिक पदार्थ ( DNA is a actually genetic material) है।

Griffith experiment: an introduction.

It may come as a surprise that less than a century ago, even the most educated members of the scientific community were unaware that DNA was a hereditary material. Frederick Griffith conducted a series of experiments with Streptococcus pneumonia bacteria and mice in 1928 and concluded that the R-strain bacteria must have picked up a "transforming principle" from the heat-killed S bacteria, allowing them to "transform" into smooth-coated bacteria and become virulent.

In this article, we'll look at one of the classic experiments that led to the discovery of DNA as a genetic information carrier.

Who was Frederick Griffith?

The "Griffith's Experiment," carried out by English bacteriologist Frederick Griffith in 1928, described the transformation of a non-pathogenic pneumococcal bacteria into a virulent strain.

Streptococcus pneumonia comes in several types or strains. Griffith chose two different strains for his experiment.

One strain of bacteria has smooth surfaces and is known as the smooth strain (S strain), while the other has rough surfaces and is known as the rough strain (R strain).

Bacteria of the S strain have smooth surfaces because they produce a polysaccharide protective coating that forms the outermost layer.

Apart from the morphological differences, Griffith discovered another significant difference between the S and R strains of bacteria, i.e., the S strain is the "virulent" strain capable of causing death in mice, whereas the R strain is the "nonvirulent" strain that will not cause death in mice.

Griffith observed that when he injected these bacteria into mice, the mice infected with the virulent S strain died from pneumonia, whereas the mice infected with the nonvirulent R strain survived.

Griffith’s Transformation Experiment

Griffith was researching the possibility of developing a pneumonia vaccine.

He used two strains of pneumococcus (Streptococcus pneumonia) bacteria that infect mice – a virulent (causing disease) S (smooth) strain and a non-virulent type R (rough) strain.

The S strain produced a polysaccharide capsule that protected itself from the host's immune system, resulting in the host's death, whereas the R strain lacked that protective capsule and was defeated by the host's immune system.

Griffith attempted to inject mice with heat-killed S bacteria as a part of his research (i.e., S bacteria that had been heated to high temperatures, causing the cells to die). The heat-killed S bacteria, but unsurprisingly, did not cause disease in the mouse.

When harmless R bacteria were combined with harmless heat-killed S bacteria and injected into a mouse, the experiments took an unexpected turn.

Not only did the mouse develop pneumonia and die, but Griffith discovered living S bacteria in a blood sample taken from the dead mouse.

He concluded that some factor or biomolecule from the heat-killed S bacteria had entered the living R bacteria, allowing them to synthesise a polysaccharide coating and become virulent. As a result, this factor "transformed" the R bacteria into S bacteria.

Griffith called this factor the "transforming principle," concluding that it carried some genetic material from the S bacteria to the R bacteria.

This process is now known as bacterial transformation and is used in a variety of significant genetic engineering applications.

Impact of The Griffith Experiment

One of the characteristics of hereditary material is a changing phenotype . Griffith referred to the phenotypic-changing factor as the transforming principle.

His work on the transforming principle received the most attention, but only after a group of Canadian and American scientists set out to investigate the chemical nature of the transforming principle in Oswald Avery's laboratory.

Avery's group concluded in their studies that deoxyribonucleic acid was the molecule identified by Griffith as the transforming principle after conducting numerous experiments.

The implications of this discovery are farfetched because it was made at a time when scientists considered protein molecules to be genetic material.

DNA, or deoxyribonucleic acid, is now recognised as the molecule that encodes all cell functions and transmits genetic information from parent to offspring in almost every living species .

In the 1940s, however, DNA was thought to be a less qualified candidate for genetic material. Avery and colleagues' research on Griffith's experiment provided the first solid evidence that DNA could be the genetic material.

Griffith's ultimate goal was to find a way to cure pneumonia. Griffith inoculated mice with various strains of pneumococci to see if they would infect and eventually kill the mice. Griffith concluded that heat-killed virulent bacteria transformed living, non-virulent bacteria into virulent bacteria. He performed his experiment on the two strains of Streptococcus pneumonia, which differ from each other due to the presence of a polysaccharide coat.

Griffith's findings were published in the Journal of Hygiene. In 1928, his experiments with mice led to his major discovery of bacterial transformation. Griffith's experiment discovered that bacteria can transfer genetic information through transformation.

Conjugation is the primary mechanism for the spread of antibiotic resistance in bacteria, and it is critical in the evolution of the bacteria that degrade novel compounds such as pesticides created by humans, as well as in the evolution, maintenance, and transmission of virulence.

The advent of artificial intelligence (AI) presents several new and exciting opportunities for improving the quality of education. While several ways of integrating AI into schooling have been explored, only a few of them consider changing the traditional school operations and educational practices.

In a recent article that was published on 23 July 2024 in the ECNU Review of Education , Professor Yong Zhao from the University of Kansas explored more radical changes that may be applied to traditional schooling to fully utilize the potential of AI technology for benefiting students.

"Numerous publications have appeared, all trying to suggest, recommend, and predict the future of AI uses in education. However, most of the discussions, regardless of their scholarly quality, are primarily focused on using AI in the traditional arrangement of schools," explains Professor Zhao.

"The assumption is that everything the traditional school has operated with shall remain the same. AI tools, according to most of the advice, are to be incorporated into teaching by teachers just like previous technologies."

By investigating a broader view of possible changes that can be applied to the current schooling system, the article not only suggested ways of utilizing the potential of AI technology better but also dealt with leveraging AI to support personalized learning. The article notes that although personalized learning has clearly demonstrated benefits, it has not been widely implemented in schools in its true sense. AI tools present a unique opportunity to implement personalized learning, customized to individual student needs harnessing their unique talents and potential.

Traditional schooling systems aim to create members of the workforce. However, AI has disrupted the job market, eliminated traditional career roles and created new roles. In the article, Professor Zhao noted that focusing and building on each child's innate talents and unique strengths is essential for them to be successful in any career of their choice. Since any potential talent when sufficiently honed is valuable in the age of AI, he argued that educational systems must focus on students' strengths rather than their weaknesses.

Furthermore, the article suggested that the traditional curricula might need to change to make way for personalized education. Students could use AI and other resources to follow their interests and passion. This might also eliminate the requirement for age-based classes and promote learning with tools, resources, experts and peers aligned with the same interests rather than age.

Besides personalized education, AI can also be effective in facilitating project-based learning. The article noted that AI tools can help schools inculcate skills such as problem solving and independent thinking in students, transforming them into individuals with critical thinking and analytical skills.

Integrating AI into the education system will also transform the role of teachers. They would become coaches and mentors who would work with the students to help them identify their strengths and potential and guide them to become the best versions of themselves. They would also need to be updated with the AI tools and help students utilize AI as a learning partner.

Traditional educational systems resist change, and now, with the advent of AI, there are several more incentives for changing how schools operate. In the article, Professor Zhao examines the question of whether and how educational systems can change in the age of AI. By considering broad-based changes to the schooling system, he suggests that the true potential of AI in learning can be unlocked.

"AI is no doubt a powerful technology, but it is easy to underestimate its power. Uses in the traditional classroom to assist students and teachers in learning and teaching helps, but they also minimize the transformative power of AI," Professor Zhao observes.

"Schools could be transformed with the advancement of technology , especially generative AI. The changes should start with student -driven personalized learning and problem-oriented pedagogy," he concludes.
